In a pharmaceutical suspension, insoluble particles are dispersed in a liquid. Those particles can remain relatively separate (deflocculated) or form loose, reversible aggregates called flocs (flocculated).

The counterintuitive point is that faster settling is not always worse. A well-flocculated suspension may settle more quickly but form a loose sediment that is easier to redisperse, while a deflocculated suspension may settle slowly yet eventually form a hard cake.

Why formulation is a balancing act

An ideal suspension should remain uniform long enough to measure a dose and should redisperse easily after storage. Formulators therefore control particle size, zeta potential, viscosity, flocculating agents and structured vehicles.

The goal is not simply “stop settling.” Complete prevention of settling is often unrealistic.

Controlled flocculation

In controlled flocculation, formulators deliberately adjust interparticle forces so particles form weak networks rather than dense sediments. Electrolytes, polymers or surfactants can influence this behaviour.
